Mechanism(s) for age-related sex differences in diet-induced cardiomyopathy: role of RNA methylation
نویسندگان
چکیده
Abstract Background/Introduction Age and sexual dimorphism contribute to the differential cardiometabolic dysfunctions associated with diet-induced obesity. However, underlying mechanisms remain elusive. RNA modifications via m6A methylation, is an emerging mediator of stability, translation localization responsible for regulation multiple biological functions. are regulated by “writers”, “erasers” “readers”. The role methylation machinery genes in heart largely unexplored, may provide insight into influence age sex on dysfunction. Purpose We aim determine changes within a cardiomyopathy mouse model, stratified according sex. Methods Male female C57BL/6 mice (6–8wk-old) were fed normal chow (NC) or high-fat/high-sucrose (HFHS) diet 1 4 months. Echocardiographic measurements performed at months American Society Echocardiography European Association Cardiovascular Imaging guidelines. At study endpoint, glucose insulin tolerance testing was conducted injecting intraperitoneally 2g/kg 0.6U/kg insulin, monitoring blood levels over 2 hour period. from tissue subjected quantitative PCR (FTO, ALKBH5, METTL3, METTL4, METTL14, YTHDF1 YTHDF2). PPIA used normalise qPCR data. Results Both male showed evidence age- metabolic dysfunction, however, males females markedly different impairments. For example, exacerbated HFHS but not females; only impaired tolerance. that had systolic (stoke volume, cardiac output) diastolic (E/A ratio) dysfunction after diet, while unperturbed. identified METTL3 “writers” consistently increased hearts vs unchanged decreased females, irrespective diet. Conversely, YTHDF1, “reader”, vs. significantly hearts. Conclusion Our confirms impairments occur versus response long-term Despite significant impairment both sexes, evident males. differentially expressed sex, suggesting be involved age-related Funding Acknowledgement Type funding sources: Foundation. Sex Differences in Age-Related Cardiovascular Mortality
INTRODUCTION Sex-related physiological differences result in different expressions of diseases for men and women. Data are contradicting regarding the increase in the female risk for cardiovascular disease (CVD) at mid-life. Thus, we studied possible sex differences in age-adjusted mortality for CVD and non-vascular diseases stratifying our findings by specific age groups. METHODS Over one mi...
متن کاملage-related differences in postural control: role of sensory information
postural control requires the integration of sensory information in the visual, vestibular, and senses the body systems. also research has shown that the sensorysystems involved in postural control, decreased with increasing age. the aim ofthis study was to investigate the effects of age and manipulation of sensoryinformation on the postural control. for this purpose, displacement of the anterio...
متن کاملPrior exercise training attenuated Doxorubicin-induced hepatic oxidative stress in Wistar rats: Age-related differences
Introduction: Previous studies have confirmed the sidelong effects of Doxorubicin (DOX) on healthy tissues, but pretreatment effect of regular Aerobic exercise in restrain of hepatotoxicity induced by doxorubicin is not clear. The aim of this study was to investigate the pretreatment effect of regular aerobic exercise on hepatic oxidative stress induced with doxorubicin of Wistar rats in differ...
متن کاملHand Function Related to Age and Sex
Objectives: The purpose of this study was for evaluation of relationship between hand function and age, sex by Jebson Taylor Test. This study determined whether age might be a factor when associated with sex. Methods: In this study Jebson Taylor was evaluated on hand in 180 normal males and females (0.50% male, 0.50% female) in each of the following 7 subtest in 3 age groups: (15-25), (35-45)...
متن کاملSex- and age-related differences in bilirubin concentrations in serum.
The relation between age and sex and the concentration of bilirubin in serum was evaluated in 6740 men and 11 215 women, ages 13 to 96 years. Mean serum bilirubin concentrations in the men significantly exceeded values in the women over all age groups examined. Further, mean serum bilirubin concentrations were greatest both in males and females in the 19-24 years age group and then declined to ...
